Infection with Shiga toxin-producing Escherichia coli (STEC) has emerged as an important cause of hemorrhagic colitis and the severe complication hemolytic uremic syndrome (HUS). STEC strains produce two structurally related toxins: Stx1 and Stx2, which bind to and cause damage to target cells, principally renal endothelial cells, by inhibiting the activity of V-ATPase, a proton pump necessary for intracellular acidification. This results in the impairment of protein and DNA synthesis, induction of apoptosis, and the subsequent vascular collapse characteristic of HUS. It is becoming increasingly apparent that the beneficial aspects of bacterial cell wall constituents can be used to improve host defense against pathogens, with the rational design of a new class of adjuvants called mucosal adjuvants. In this chapter, we describe methods for the purification of Stx1 and Stx2 and evaluate their immunogenicity and adjuvant properties in vivo, using mice as a model. We also describe methods for their delivery by the nasal, pulmonary, and oral routes, as well as the detection of the toxin in blood and tissues using specific antibodies and/or enzyme-linked immunosorbent assay methods.
